Multimerization and Reconfiguration of Preformed DNA nanostructures through Toehold-Mediated Strand Displacement

Abstract

Structural DNA nanotechnology has advanced at an extraordinary pace in the past three decades. Increasingly, more complex structures have been demonstrated in the field, utilizing three major categories of DNA nanostructures— the branched DNA motif, the DNA origami and the single-stranded tiles. Overall, they have been developed as very handy tools to image, target and deliver in biological and medical applications. However, the size and the complexity of these DNA nanostructures are the limiting factors when they are applied in more complex systems and fields. Here, we polymerize and reconfigure the DNA nanostructures for more complex applications, by using the mobile four-way toehold-mediated displacement, such that, with the help of the single- stranded tiles without scaffold strands, the resulting DNA nanostructures would be higher ordered and more complex for further applications.
